Focused Ultrasound Brain Drug Delivery System: Design and Development

Focused ultrasound (FUS) technology, which appears half century ago, has got more and more attention on its clinical therapeutic potential in recent years. But only within few years, focused ultrasound with the presence of microbubbles has been noticed to be capable of locally and reversibly disrupting blood-brain barrier (BBB) without causing neuronal damage and proof its potential in the delivery of macromolecules (> 400 Da) into CNS, including those therapeutic agents (> 90% among all) for CNS therapy which may not naturally cross the barrier into brain, and brings bright hopes for noninvasive brain drug delivery for treating brain cancers or neurodegenerative diseases. A phased array ultrasound system is essential for FUS-BBB opening to enable electronic steering and correction of the focal beam which is distorted by cranial bone. In this presentation, we demonstrate our development experience of multiple-channel ultrasound phased array system for large-region transcranial BBB opening in the brains of large animals. We also demonstrated the capability of exposure from ultrasound phased array system to potentially enhance the BBB-opening effect.
